Giant african snail, Achatina fulica (Férussac, 1821): an environmental and public health problem in the northwestern of Paraná State, Brazil Biological Sciences
Andreazzi, Márcia Aparecida; Gasparotto, Francielli; Paccola, Edneia Aparecida de Souza; Silva, Cleiltan Novais da; Rodrigues, Aparecida de Fatima Cracco; Lizama, Maria de los Angeles Perez.
Achatina fulica, known as Giant African Snail (GAS), was introduced in Brazil as a substitute for the European escargot, Helix aspersa maxima. However, its cultivation was abandoned and the mollusk became an invasive species, found throughout Brazil and causing damage to health and the environment. In this context, this study analyzed the knowledge of the population about the damage caused by GAS, seeking to reveal a regional scenario on this issue. This exploratory and cross-sectional study was conducted by means of a questionnaire applied to 150 people, which obtained sociodemographic characteristics of respondents and specific responses about the knowledge and handling of GAS. The results showed that most of the respondents know the snail and are...

Development of quality index method for eviscerated and non-eviscerated octopus (Octopus insularis) Rev. Ciênc. Agron.
Aragão,Márcia Facundo; Garruti,Deborah dos Santos; Ogawa,Norma Barreto Perdigão; Bezerra,Viviane Costa; Silva,Elisabeth Mary Cunha da.
ABSTRACT This work aimed to develop a Quality Index Method (QIM) for octopus species Octopus insularis and apply it to samples of eviscerated and non-eviscerated specimens in order to establish stability in cold storage. Initially we carried out the recruitment of judges, who were trained to verify the changes undergone by the octopus during storage under refrigeration. The development of QIM Table took place in three experiments. At first, a QIM Table for Octopus vulgaris from literature was presented to judges together with specimens of O. insularis, so they could assess whether the parameters were suited to this specie. In the second experiment the objective was to validate the table, verifying if changes were needed in the predefined parameters or if...

Spatial distribution of mollusks in the intertidal zone of sheltered beaches in southeastern of Brazil Zoologia
Arruda,Eliane P. de; Amaral,A. Cecilia Z..
The spatial distribution of mollusks in the intertidal zone was examined monthly from August 1995 through July 1997, in Enseada, Barra Velha and Araçá beaches in southeastern of Brazil. One study sector was selected in Enseada and Barra Velha, and two sectors in Araçá (Araçá I and Araçá II). The sectors were 10 m wide and equivalent in length to the width of the intertidal zone. Each sector was divided into three horizontal levels: lower, middle and upper, where the samples were taken with a cylinder corer with a base area of 0.16 m². In order to characterize the intertidal environment in these areas, some environmental variables were analyzed. In general, the mollusks were distributed in the sectors as follows: Enseada - Olivella minuta (Link, 1807) in...

The new insights into the oyster antimicrobial defense: Cellular, molecular and genetic view ArchiMer
Bachere, Evelyne; Rosa, Rafael Diego; Schmitt, Paulina; Poirier, Aurore; Merou, Nicolas; Charriere, Guillaume; Destoumieux-garzon, Delphine.
Oysters are sessile filter feeders that live in close association with abundant and diverse communities of microorganisms that form the oyster microbiota. In such an association, cellular and molecular mechanisms have evolved to maintain oyster homeostasis upon stressful conditions including infection and changing environments. We give here cellular and molecular insights into the Crassostrea gigas antimicrobial defense system with focus on antimicrobial peptides and proteins (AMPs). This review highlights the central role of the hemocytes in the modulation and control of oyster antimicrobial response. As vehicles for AMPs and other antimicrobial effectors, including reactive oxygen species (ROS), and together with epithelia, hemocytes provide the oyster...

Characterization of GnRH-related peptides from the Pacific oyster Crassostrea gigas ArchiMer
Bigot, Laetitia; Zatylny-gaudin, Celine; Rodet, Franck; Bernay, Benoit; Boudry, Pierre; Favrel, Pascal.
Gonadotropin-releasing hormone (GnRH), a key neuropeptide regulating reproduction in vertebrates has now been characterized in a number of non-vertebrate species. Despite the demonstration of its ancestral origin, the structure and the function of this family of peptides remain poorly known in species as distant as lophotrochozoans. In this study, two GnRH-related peptides (Cg-GnRH-a and CgGnRH-G) were characterized by mass spectrometry from extracts of the visceral ganglia of the Pacific oyster Crassostrea gigas. These peptides showed a high degree of sequence identity with GnRHs of other mollusks and annelids and to a lesser extent with those of vertebrates or with AKH and corazonins of insects. Both the mature peptides and the transcript encoding the...

Intracellular microcolonies of bacteria (IMC), in some cases developing large extracellular cysts (bacterial aggregates), infecting primarily gill and digestive gland, have been historically reported in a wide diversity of economically important mollusk species worldwide, sometimes associated with severe lesions and mass mortality events. As an effort to characterize those organisms, traditionally named as Rickettsia or Chlamydia-like organisms, 1950 specimens comprising 22 mollusk species were collected over 10 countries and after histology examination, a selection of 99 samples involving 20 species were subjected to 16S rRNA gene amplicon sequencing. Phylogenetic analysis showed Endozoicomonadaceae sequences in all the mollusk species analyzed....

One Case of Triphallia in the Marine Snail Echinolittorina peruviana (Caenogastropoda: Littorinidae) International Journal of Morphology
Castillo,Viviana M; Brown,Donald I.
Gonochoric representatives of Littorinidae have a reproductive system organized with transit organs that connects the testicle with a penis specialized for internal fertilization. However, malformations at this level can produce, like triphallia (presence of triple penis). This description corresponds to one case of triphallia found in Echinolittorina peruviana (Lamarck, 1822) inhabitant of the South Pacific coasts. The macro and microscopic analysis reveals that every penis is an independent appendix whose individual morphology is similar to that in normal individuals, characterized by a non pigmented conic penis associated with a mammilliform gland located at the base of the right ocular tentacle. The cervical sperm groove lined by a simple ciliated and...

Microscopic Anatomy of the Reproductive System in Two Sympatric Species of Fissurella Bruguiére, 1789 (Mollusca: Vetigastropoda) International Journal of Morphology
Collado,Gonzalo A; Brown,Donald I.
This study describes the microscopic anatomy of the reproductive systems of the keyhole limpets Fissurella latimarginata and Fissurella cumingi to test the existence of a possible isolating reproductive mechanism of morphophysiological type between species, and consequently to determine if the findings provide a valid taxonomic characteristic to distinguish these sympatric species. The results show that in both species the reproductive system is intimately associated with the excretory system. The gonad has a septa organization, and is connected with the distal portion of the right kidney by means of a renopericardial duct which functions as a genital duct. The gametes are evacuated to the mantle cavity through the right kidney opening. Besides transport...

Since 2008, juvenile Crassostrea gigas oysters have suffered from massive mortalities in European farming areas. This disease of complex etiology is still incompletely understood. Triggered by an elevated seawater temperature, it has been associated to infections by a herpes virus named OsHV-1 as well as pathogenic vibrios of the Splendidus clade. Ruling out the complexity of the disease, most of our current knowledge has been acquired in controlled experiments. Among the many unsolved questions, it is still ignored what role immunity plays in the capacity oysters have to survive an infectious episode. Here we show that juvenile oysters susceptible to the disease mount an inefficient immune response associated with microbial permissiveness and death. We...

Use of OmpU porins for attachment and invasion of Crassostrea gigas immune cells by the oyster pathogen Vibrio splendidus ArchiMer
Duperthuy, Marylise; Schmitt, Paulina; Garzon, Edwin; Caro, Audrey; Rosa, Rafael D.; Le Roux, Frederique; Lautredou-audouy, Nicole; Got, Patrice; Romestand, Bernard; De Lorgeril, Julien; Kieffer-jaquinod, Sylvie; Bachere, Evelyne; Destoumieux-garzon, Delphine.
OmpU porins are increasingly recognized as key determinants of pathogenic host Vibrio interactions. Although mechanisms remain incompletely understood, various species, including the human pathogen Vibrio cholera, require OmpU for host colonization and virulence. We have shown previously that OmpU is essential for virulence in the oyster pathogen Vibrio splendidus LGP32. Here, we showed that V. splendidus LGP32 invades the oyster immune cells, the hemocytes, through subversion of host-cell actin cytoskeleton. In this process, OmpU serves as an adhesin/invasin required for beta-integrin recognition and host cell invasion. Furthermore, the major protein of oyster plasma, the extracellular superoxide dismutase Cg-EcSOD, is used as an opsonin mediating the...

Acumulación de ácido araquidónico en fosfolípidos y triglicéridos y producción de prostaglandinas con relación a estrés térmico en semilla de ostión Crassostrea gigas ArchiMer
Duran Encinas, Yazmin.
Crassostrea gigas is a marine invertebrate that inhabits the intertidal zone and therefore, is commonly exposed to multiple environmental stressors, including temperature. In recent years mass mortality events have been reported in Mexico in C. gigas associated with the increase in temperature during the summer. Organisms under thermal stress present an increase in prostaglandins (PG) production by cyclooxygenases (COX), an enzymatic route using arachidonic acid (ARA) present in the phospholipids (FL) of the cell membrane as substrate, but PG can also increase non-enzymatically by reactive oxygen species (ROS) that are produced during increased metabolism. It has been reported that some organisms exposed to stress accumulate a higher concentration of ARA...

Epigenetic inheritance and intergenerational effects in mollusks ArchiMer
Fallet, Manon; Luquet, Emilien; David, Patrice; Cosseau, Céline.
Recent insights in evolutionary biology have shed light on epigenetic variation that interacts with genetic variation to convey heritable information. An important characteristic of epigenetic changes is that they can be produced in response to environmental cues and passed on to later generations, potentially facilitating later genetic adaptation. While our understanding of epigenetic mechanisms in vertebrates is rapidly growing, our knowledge about invertebrates remains lower, or is restricted to model organisms. Mollusks in particular, are a large group of invertebrates, with several species important for ecosystem function, human economy and health. In this review, we attempt to summarize the literature on epigenetic and intergenerational studies in...

Temporal and spatial variation on heavy metal concentrations in the bivalve Perna perna (LINNAEUS, 1758) on the northern coast of Rio de Janeiro State, Brazil BABT
Ferreira,Alexandre Gomes; Machado,André Luís dos Santos; Zalmon,Ilana Rosental.
Heavy metal (Cd, Cr, Cu, Fe, Mn, Ni, Pb and Zn) concentrations were studied in Perna perna from three beaches (Barra do Furado, Buena and Ponta do Retiro) on the northern coast of Rio de Janeiro State by ICP-AES. The average concentration was 0.3 ± 0.04 (Cd); 1.9 ± 0.6 (Cr); 6.1 ± 0.7 (Cu); 1,130 ± 113 (Fe); 22 ± 3.2 (Mn); 9.3 ± 4.6 (Ni); 0.4 ± 0.2 (Pb); 44 ± 5.8 (Zn) µg.g-1 dry weight. There were no significant difference for almost all the studied metals in relation to sex. Although, significant spatial variations (p < 0.05) occurred for Cr, Pb and Ni with higher values for Barra do Furado. Temporal variations were significant (p < 0.05) at the three beaches, however, no trends of accumulation were observed. The metal concentrations were similar to...

Evidence of a bactericidal permeability increasing protein in an invertebrate, the Crassostrea gigas Cg-BPI ArchiMer

A cDNA sequence with homologies to members of the LPS-binding protein and bactericidal/permeability-increasing protein (BPI) family was identified in the oyster Crassostrea gigas. The recombinant protein was found to bind LIPS, to display bactericidal activity against Escherichia coli, and to increase the permeability of the bacterial cytoplasmic membrane. This indicated that it is a BPI rather than an LPS-binding protein. By in situ hybridization, the expression of the C gigas BPI (Cg-bpi) was found to be induced in hemocytes after oyster bacterial challenge and to be constitutive in various epithelia of unchallenged oysters. Thus, Cg-bpi transcripts were detected in the epithelial cells of tissues/organs in contact with the external environment (mantle,...

Oyster hemocytes express a proline-rich peptide displaying synergistic antimicrobial activity with a defensin ArchiMer
Gueguen, Yannick; Romestand, Bernard; Fievet, Julie; Schmitt, Paulina; Destoumieux Garzon, Delphine; Franck, Vandenbulcke; Philippe, Bulet; Bachere, Evelyne.
A cDNA sequence that encodes a 61-amino acid polypeptide precursor with homologies to proline-rich antimicrobial peptides (AMPs) was identified in the oyster Crassostrea gigas. After release of a hydrophobic signal peptide. the resulting 37-amino acid peptide, Cg-Prp, is composed of an acidic region and a cationic proline-rich region. To evaluate the biological properties of Cg-Prp, multiple proline-rich peptides corresponding to putative processing of the full-length Cg-Prp were synthesized. A limited antimicrobial activity was observed for two of them, which also showed strong synergistic antimicrobial activity with Cg-Def, a defensin from C gigas. To our knowledge, this is the first evidence of synergy between a defensin and another AMP in an...

Adult somatic progenitor cells and hematopoiesis in oysters ArchiMer
Jemaa, Mohamed; Morin, Nathalie; Cavelier, Patricia; Cau, Julien; Strub, Jean Marc; Delsert, Claude.
Long-lived animals show a non-observable age-related decline in immune defense, which is provided by blood cells that derive from self-renewing stem cells. The oldest living animals are bivalves. Yet, the origin of hemocytes, the cells involved in innate immunity, is unknown in bivalves and current knowledge about mollusk adult somatic stem cells is scarce. Here we identify a population of adult somatic precursor cells and show their differentiation into hemocytes. Oyster gill contains an as yet unreported irregularly folded structure (IFS) with stem-like cells bathing into the hemolymph. BrdU labeling revealed that the stem-like cells in the gill epithelium and in the nearby hemolymph replicate DNA. Proliferation of this cell population was further...

The effect of stocking density on the growth of apple snails native Pomacea bridgesii and exotic Pomacea lineata (Mollusca, Gastropoda) Anais da ABC (AABC)
JUNIOR,ESMAR SOUZA; BARROS,JOSE CARLOS N. DE; PARESQUE,KARLA; FREITAS,RODRIGO R. DE.
The demand for alternative food sources is currently in evidence in the world and, therefore, the culture of animal species considered not conventional makes this theme relevant and appropriate. In the present study, the species Pomacea lineata and Pomacea bridgesii, each with three stowage densities (0.5 [T1], 1 [T2], and 1.5 [T3] animal/L), were tested. They were analyzed regarding growth rate, weight gain, final biomass, feed conversion and percentage of survival. There was not any statistically significant difference between the different densities for both species. The final average weight in the three waterworks did not differ significantly in P. bridgesii. In P. lineata, T1 (22.3 ± 1.80g) was significantly higher than T2 and T3. On the other hand,...
